Isn't 15 1/2 the legal age to get your learner's permit?  I don't know.  I didn't even try driving a car until I was 23 (personal choice not to and was probably wise I didn't since I was on the immature side) .  My brother got his learners somewhere around 15 1/2 and got his liscence when he turned 16, on his 16th birthday.

Does your nephew have autism or aspergers?   Even if he did have autism or aspergers, if he's capable of driving, then his parents should let him  drive and take the precautions every parent does with their child.  Are you involved in the decision making for your nephew?  If you have concerns voice them to your sister, but in the end if it's her son and she's the decision maker, you have to accept what she decides for her own child and not harrass her about her choices if you don't agree with them.  Sorry about the lecture... am dealing with relatives who have been harrassing me about what I can and can't do, so I guess it's a sore topic for me.

What my parents did with my brother since he so desperately wanted to drive ASAP, was they enrolled him in the high school driver's ed behind the wheel course.  After that ended, they enrolled him in a private driving school.  They wouldn't let them take their car out except driving with them in an empty parking lot with cones.

I have a friend who has been driving since she was 10.  She grew up on a farm in Iowa.  She said her dad had her drive (alone) to go get her mom from an airport in a blizzard.  Of course, she said she was driving illegally since she wasn't even old enough for the license driving alone.  Personally I think her parents were NUTS!  but I guess it was their choice and if anything would have happened, they would have had to live with the consequences.
