Can you get an inaccurate reading checking for brain cancer if wig isn't removed ?

Hi.  If you're talking about a head CT scan, the x-rays used for the scan can readily penetrate through the wig.  So it would not matter if you're wearing a wig  when the scan is done.  What would affect the quality of the CT scan are movements of the head, but there are effective means of immobilizing the head before the procedure.
