need help to undestand

My son goes to pre-school 2 full days and 3 half days he is 4 yrs. old.
One time my son said , he doesn't want to go to school nobody plays with me , i don't like going to school .... two days in a row he said the same thing. I got a bit concernd so i asked his teacher is everything ok she said that one of his class-mate (whom my son plays with him everytime ) is not here, so he is quite in school and doesn' play that much. I explained him try to make new friends so that he can play with others too.
Last week my son's teacher is out for weeks for some personal reasons, so there are new teachers coming at different time. When I went to pick him he was crying too much, i thought he saw a bad dream during his nap time so maybe he is crying, then I saw the class-mate (whom he plays more) is not there for some reason he left early during the nap time,then I thought my son got scared because my son didn't see him after he woke up.
Now he is not willing to go to school giving lots of reasons like "I don't like to go to school", " I'm scared", "I'm scared of my teacher " , " nobody plays with me" etc....
My son asks every time whether i'm going to pick him up ... he is getting more insecure whether i'm going to pick him up.
Today i went to school heard from teacher he was screaming and crying not willing anybody to touch him or anything going here and there crying a lot later some senoir teacher calm him down, but still he was crying, the substitute teacher is telling that she is upset saying that is she the reason that my son is crying or does my son have some psychological problems he is crying..
I' m really worried why my son is doing this , he insecure because his old teacher is not there or so, I tried to ask him why is the reason he is crying or why he is insecure of picking up but everytime he is telling something different, But when he is home his behavior good as he used to behave.
I never said him about not picking up or so as he is getting insecure about asking me everytime whether I'm going to pickup him from school.
