Birinapant to take or not!!

On internet there,s a website that is supplying birinapant in India. However they asked me a lot of questions like what form do I need it, injectable powder cap tab etc, what purpose I need it, what amount I need it? I simply answered whats the price of per unit but they weren't clear about the unit neither me. This made me confused what to order for as there is no clear picture about the drug. I am here to ask weather it's advisable to take it without any guidance by a doctor or researcher. If yes then whats the way ie to take injectable or oral and in what quantity per day till how long and how much. What to order for(vial or powder)? Is the source of supply Is authentic? Is the palsy caused by it is reversible or irreversible? Can it cause permanent brain injury or other vital organ damage? Whats the safe dose? Though this topic was discussed earlier once but I thought that a fresh perspective should be taken at it since there is no curing drug coming and this makes pave own ways in desperation.

I wouldn't use it. Sadly it may take another 5-10 years before there is a cure.

The count has already begun from 2012 and its already about 5years passed sayin next 5yrs for the cure. It's heartbreaking

No, Hbv isn't a "harmless" disease and the antivirals we use has caused me more side effects than anything. When you've known or had a friend die of Hbv I wouldn't say it's "harmless".

thanx friends for ur helpful replies. steff u r right justifying virus as harmless and study for hope is very correct on effect on mouse and human can be different since hbv is a human virus. Thanx stephen for eye opening. The Wait continues.

The amount of acidity introduced during the 30 min infusion is too small to cause any side effects.  Actually the birinapant infusions do not cause any immediate side effects. Problems are rather related to the proapoptotic effect itself that is not limited to the liver, as expected. Typically the pancreatic lipase increases, but not to dangerous levels. It is unclear if beta cells are effected also. At longer use podocyte function needs to be carefully monitored following urine  microalbumin levels.

I too was very excited when the press release first came out, reinforced by the fact it was from a very prestigious Australian institute. It was news repeated all over the Internet for days. With rapid advance into a phase 2 clinical trial, again in Australia, our hopes were all raised. Then came the suspension of the trial, followed by deafening silence. I wrote to the public relation dept of WEHI, he washed his hands over the whole affair and passed responsibility to Tetralogic, even though their chief investigator was an advisor to Testralogic, the same one who flowed the idea that there will be a new clinical trial in India, in July 2016.
I learned a painful lesson and is now more cynical.

I read the following at the website of Tetralogic regarding their clinical trials:

"In July 2015, we announced that we intend to initiate a combination single ascending dose/multiple ascending dose clinical trial, with birinapant as a single agent, in chronic HBV subjects. These subjects will not be taking other antiviral medication. In the single ascending dose phase of the trial, subjects will be given a single dose of birinapant. The dose of birinapant will be escalated until at least one of the subjects shows evidence of activity, defined as an increase in liver transaminases and/or a decline in circulating viral DNA. At that point the cohort will be expanded and additional subjects will each receive four weekly administrations, at that dose, of birinapant. The starting dose of birinapant will be 2.8 mg/m2. We have retained a clinical research organization and currently expect to initiate this trial at multiple sites in India in early 2016. Timing of results will depend upon enrollment rates and upon the cohort in which activity, if any, is seen."
But from your post I kind of read these trials are terminated, is that true?
The old clinical trial, Entecavir + Birinapant, was stopped because of side effect. The new trial, planned to be in India in 2016, has not started. Personally, I am doubtful that there will be any new Birinapant trial for HBV in the near future, mainly because Tetralogic is a very small company and will need to preserve fund to continue their clinical trial of Birinapant in the treatment for cancer.
My gripe is with the original press release from Walter Eliza Hall Institute:
"Australian scientists have found a potential cure for hepatitis B virus (HBV) infections, with a promising new treatment proving 100 per cent successful in eliminating the infection in preclinical models."
They could have warned that the preclinical models involved mice that had to be engineered to be infected with HBV, and that killing infected liver cells in human can be very dangerous.

Birinapant cannot meaningfully used orally, there is almost no absorption from the gi tract. It has to be infused over 30 minutes from a solution of pH 5, 50ml approximately, in  50 millimolar of sodium citrate once weekly at a dose of no more than 15mg per square meter. This is the way it is used in the cancer trials.

you can easily end up dead with this vs an hameless virus like hbv.dont waste your time or life for nothing

stef2011, do you really think hbv is a harmless virus? According to me who is sufferign from chronic hbv, i believe this virus is indeed harmfull and dangerous..
with all the antivirals we have it is nothing

There is absolutely no proof that Birinapant will cure HBV. Don't believe in the hype expressed in press release from Walter Eliza Hall, the institute where the scientists work.

You are correct, there is no trace of evidence published that birinapant can cure hepatitis b in humans. The hype was supported by the impressive animal  results and the fascinating theoretical  concept to eradicate cells carrying the cccDNA or even integrated hbv genomic segements.

We have discussed tetralogic and other aspects of birinapant use here several times.

I have lately also posted about some hidden dangers of prolonged use.
