Abstract
The hypolipidemic effects of pantothenic acid derivatives (phosphopantothenate, panthenol and pantethine) were studied in mice with hypothalamic obesity. Hypothalamic obesity in mice was induced by single injection of aurothioglucose (300 mg/kg body wt, i.p.). All the tested substances were administered during the last 10 days before decapitation (i.m., of dosage equivalent to 150 mg/kg body wt of phosphopantothenate). The studied substances inhibited the weight gain of the animals with hypothalamic obesity over the last 10 days of the experiment. The treatment with aurothioglucose increased food intake and mean body weight, blood glucose level; insulin, serum total cholesterol, triglyceride, the sum of LDL + VLDL and LDL-cholesterol concentration; triglyceride and cholesterol fractions in the liver; triglyceride and FFA content as well as lipoprotein lipase activity in adipose tissue of experimental mice. The administration of the assay compounds lowered food intake and mean body weight, insulin and glucose levels and decreased the content of triglycerides, total cholesterol and cholesterol esters in serum and adipose tissue as well as raised the activity of lipoprotein lipase in adipose tissue and serum lipolytic activity in obese mice. Among the compounds studied the reverse effect of panthenol was especially pronounced. The mechanism of hypolipidemic effects of pantothenic acid derivatives can be related to the reduced resistance to insulin and activation of lipolysis in serum and adipose tissue.

Combined pantethine and probucol therapy for Japanese patients with non-alcoholic steatohepatitis.

Abstract
Aims: To evaluate the efficacy and mechanism of combined pantethine and probucol therapy in Japanese patients with non-alcoholic steatohepatitis (NASH), liver function, serum cytokines, serum adiponectin and liver biopsy findings were investigated. Methods: Sixteen patients with NASH and hyperlipidemia were treated with pantethine (600 mg/day) and probucol (500 mg/day) for 48 weeks. Results: The mean pretreatment aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels were 66 IU/L and 113 IU/L, respectively, which showed a significant decrease with treatment to 33 IU/L and 51 IU/L (P < 0.01 and P < 0.05), respectively. Total cholesterol was also significantlydecreased (P < 0.01). In addition, the mean serum TGF-beta level was significantly decreased, while the mean serum level of high molecular adiponectin was increased. In eight patients, liver biopsy was performed both before and after treatment. In four of these patients, inflammation was improved, and fibrosis was improved in two patients. No side-effects of this treatment were noted. Conclusion: Combined pantethine and probucol therapy was safe and effective for Japanese NASH patients, with its efficacy being mediated through adiponectin and TGF-beta.

Abstract
Pantethine, the stable disulfide form of pantetheine, is the major precursor of coenzyme A, which plays a central role in the metabolism of lipids and carbohydrates. Coenzyme A is a cofactor in over 70 enzymatic pathways, including fatty acid oxidation, carbohydrate metabolism, pyruvate degradation, amino acid catabolism, haem synthesis, acetylcholine synthesis, phase II detoxification, acetylation, etc. Pantethine has beneficial effects in vascular disease, it able to decrease the hyperlipidaemia, moderate the platelet function and prevent the lipid-peroxidation. Moreover its neuro-endocrinological regulating role, its good influence on cataract and cystinosis are also proved. This molecule is a well-tolerated therapeutic agent; the frequency of its side-effect is very low and mild. Based on these preclinical and clinical data, it could be recommended using this compound as adjuvant therapy.
