I worked in microbiology research for several years but left the field in 1986.  New test methods have come along as we find new diseases and improve upon previous test methods.  I am going to send you a link from the web site, Lab Test Online, which tells about Antibody tests.  I hope this will give you a basic understanding of why we use antibody development as one form of diagnostic testing to see if a patient has a particular disease.  http://labtestsonline.org/understanding/analytes/antibody_tests/glance.html
This site is sponsored by the American Society of Clinical Pathologists, the group that licenses medical technologists, such as myself.  You will see the HON code at the bottom of the page.  My second degree is as a medical librarian and we like to recommend sites that display this icon.  As you know, anybody can put together a web site, and this is a good frame of reference that you have found a reputable site.  This is a short portion of the information retrieved from this article, "Some antibody testing focuses on specific IgM, IgG, IgA and/or IgE tests. IgG and IgM tests are used primarily for diagnosis and monitoring of infectious diseases or for determination of immune status."
Here is the link to the section that is specific to Lyme disease:  http://labtestsonline.org/understanding/analytes/lyme/test.html

Lyme disease wasn't a parameter that we were testing for when I left research so I am in the same "boat" as you are, and trying to understand this often frustrating disease and seems to hide in our bodies and doesn't always produce the typical immune response that I learned about in school so many years ago.  I was just tested for Lyme disease on Monday and haven't heard the results yet.  Hope this helps, Kay
