Advice needed on vaginal bleeding

My wife was due to start her periods on Sunday Sept 28, but started on Saturday Sept 27.  Her period lasted for two days, Saturday was med-heavy, Sunday was very light with egg-white.  She did not bleed on the 29th or 30th, but started bleeding again yesterday (Oct 1).  Her periods usually come every 28-30 days.  This is the first time it has started on day 27 and the first time her periods started/stoped/started again.  We are also trying for a baby for the past 9 months...

Two questions:
1) Should we be concerned about this? Could pregnancy be a possibility?
2) If she is not pregnant, what day would now count as day 1 Sept 27 or Oct 1?

Many thanks
