I have 2 HIV QUESTIONS

I have 2 questions
  1.) I still don't get it is it 90 days or 12 weeks(84 days) if I take it at 12 weeks will that test be conclusive?
  2.) Can anything mess up a blood test?  Something like smoking mary jane or a taking it on a full stomach...ext.
