nonnucleoside allosteric inhibitors??

someone in here, maybe mr. liver, brought out the idea of the baffling nature of this soccer-ball-like virus. I'm starting to get the label of "baffling" ,nor any other known veriage, will quite do justice to this bug!!!

Thre are several substances in development in this class. It is hampered by very rapid development of resistance, like HCV 796 ( now terminated) , has exemplified. The reason is that the allosteric hindrance of the polymerase functioning can easily be overcome by any HCV mutation at the drugs binding site, ( that binding site is remote from the active catalytic site) and any new amino acid side chain sticking out at the binding site will reject the inhibitory drug binding, while not influencing the viral fitness/performance at the catalytic site.

The use of this class is envisioned to be of additional help to prevent resistance to the major players in a combo antiviral cocktail. In that setting it might further increase SVR percentages by blocking some of the resistance towards the other components.
