Usually a couple weeks after the unprotected sex is when you should take the test.  The boxes will tell you just how sensitive they are.

There's definitely a reason to be concerned. There's a reason you're supposed to get a shot every few months.  And yes, the shot is definitely not 100%.

Spotting could just be your body trying to adjust to not having the hormone dosage in your body any longer since you're 12 days past due on your next shot if I'm reading correctly, but it could also be cause for concern.

My mind was a day or so ahead apparently when I replied to this.  Depending on the time you had sex on Friday, you can still go get Plan B to be on the safe side.  You can take it up to 72 hours after unprotected intercourse.  If you had it Friday evening, you should still be able to take it tomorrow morning.  It can cause your period to behave strangely.

You should not be worry... the depo shot will be in your body for a year or more, it will be normal that your periods get inrregular... you can go to the ginecologist and they will give you something to get your period regular, my dr said to exersise your ovaries... lol

If it's really your period, then you're fine, but if it's different than how your periods usually are (as in just spotting or something), I would take a pregnancy test as it was possible.
