Target brain inflammation

Hi, i come from the HCV post treatment issue subforum, that board is not really fast with answers and  this goes to the people who have done Interferon treatment at least one time and who experience persistent limitations in their cognitive functioning as well as physical symtoms like fatigue , muscle weakness and so on.

the key to all these symtoms lies in the brain where Interferon acted because it is a systemic treatment. it affects the whole body, including the brain.

10 % of the brain consists of microglia. Little cells that normally sleep and get active when pathogens infiltrate the CNS for example. Interferon activates them for weeks. That makes them extremely sensitive towards noise, chemicals, food etc. and once the cascade activates themselves they start oxidative processes that attack the nerve tissue and lead to fatigue, "brain fog" etc. So, the goal is, how to calm down the microglia.

The microglia has a receptor called TLR4 or Toll Like Receptor 4. There is a number of inhibitors that block activation of microglia through TLR4. These are:

Curcumin
Dextromethorphan (DXM)
Naltrexone (LDN)
Naloxone
Thalidomide
Paclitaxel
bis-ANS
JTT705
Auranofin
Coprohemin
Amitriptyline
Cyclobenzaprine
Ibudilast
Imipramine
Ketotifen
Mianserin
Propentofylline
LPS-RS

There may be other antagonists out there, but i couldn't find them yet.

PROS AND CONS OF THE FOLLOWING TESTS:

I tested naltrexone in a low dose (3mg)

PRO: better movement coordination, less brain fog
CON: dysphoria, insomnia, lost its activity during the day

I tested DXM in a moderate dose(up to 120mg)

PRO: much better coordination, less brain fog, happiness
CON: is not good for the brain, it leads to addiction ( i read that)

I will test Curcumin (Curcuflex 47mg)

Do you have experience with TLR4 antagonists?

i am happy that i can help :-) many people think that Naltrexone is the ONLY substance that helps with brain inflammatory issues, that's not true! There are so many other substances that have an impact on this special problem and i'm going to test as many as i can get my hands on... Paclitaxel of course not
