DragonSlayer45's R7128 Trial results are in

As some of you know I have been participating in the Pharmasett Phase I polymerase inhibitor trial for R7128.   Everyone in the trial gets pegasys and copegus (SOC) and 1 in 5 get a placebo instead of the polymerase inhibitor. So this is a triple therapy. You take the trial drug for only 4 weeks along with the SOC followed by 4 weeks just SOC with the option of continuing for an additional 40 weeks SOC. I am just beginning week 7.

I know when I began this trial there were quite a few people who wanted to know how this trial was going. They trial coordinator told me after I started that I would not get my VL load results until after Day 56. Well I found out today that my Viral load is in (day 45) and it is UND (<15 IU). My baseline was greater than 6 million so this is a 6 log drop folks. They did not say at what point I cleared.

This is really exciting news for all of us who suffer from Hep C as this drug will be another future potent weapon against HCV. For those who have been following the new drugs (which is most of us) , you may recall that this drug in an earlier trial showed an 85% success rate. This part of the trial was with a lower dosage and it included geno 2 and 3 non-responders in addition to geno 1 treatment naive patients like myself. While this news in itself is very exciting, to me what is the most exciting part is that the R7128 has no sides and in earlier trials showed no evidence of Viral mutations. Unfortunately because it has to be taken, as do all of the new drugs with the SOC, I still had sides from them.

Obviously these are early results and is not an official press release and have not been reviewed and vetted by all the researchers, but I am telling you now the results from this trial will be big news when they come out officially.  I still have a way to go and will continue on for 41 more weeks with our beloved cocktail of Peg and riba but thought you would appreciate to hear the news!

That's great news!!  Congrats!!!  I was wondering how you were doing when I read the joke thread and you were cracking me up, you are too funny!  Even on tx, that must be a good thing, of course it's a drag as I've heard it isn't a cake walk for anyone, hopefully the worst part is gone and you will do better thru the rest.  God Bless

Thanks for the congrats and I totally understand where you are coming from. That is why I said this is early and it is not official but I just had to share as I did not expect this news. It is still early to tout this drug and I will let you all know when they unblind us all. I just know in my heart of hearts I did not get the placebo but you are 100% right about being cautiously optimistic.

Thanks for letting us know about how the trial is going and congratulations on being UND.  

I am going to pospone my hopes about this drug until you are told for sure that you did get it.  

I was in the Vertex Prove2 trial.  When we were unblinded we found out that one guy who was in the control group (no VX) became UND on day 15 via SOC alone.  He was geno 1 and had had hepC for over 30 years.  So maybe this sort of thing is more common than we hear about and maybe that's because unless you are in a trial you don't usually get a PCR at day 15.  

Anyway, looking forward to hearing how it pans out if you'd be kind enough to post,

Thanks for all of your well wishes. I really appreciate it. Now if I could just kick the SX of the peg and riba like I did the HCV I'll be really happy. I told the Trial coordinator that if I was to have imagined how I would have reacted to this good news (I got the call while I was driving) I would have pictured myself running around my car 10 times screaming out with joy. Instead I was happy but just really tired. I would have thought the the adrenaline rush alone of this great news would have lifted my fatigue, if even for just a minute. It did not though and that concerns me. Do not misunderstand me as I am very happy, ecstatic would be the word, but tired. I am waiting to see what the rest of my blood work shows. As of a week ago my RBC and the like was OK. These would seem to be symptoms of anemia right?

Sorry that is an excellent question and point. Although they have not unblinded the study, I would say it is safe to assume I got the trial drug and not the placebo. Considering the fact I am a male, geno 1 that has been infected for 25+ years with a high Viral load it is not likely I cleared this fast with just peg and riba. That is assumption on my part and not a fact though.
